BioGrammatics is making available on a pre-launch basis, the Pichia GlycoSwitch® SuperMan5 strains and related Contract Services. Two OCH1 mutant strains, the non-auxotrophic and his- mutant, are now available as part of a pre-launch of the Pichia GlycoSwitch® System.  The full Pichia GlycoSwitch® system will be available in the near future. **Both strains should express your target protein with a mannose-5 structure at N-linked sites. O-glycosylation may be absent or also reduced to the mannose-5 structure.

When ordering Pichia GlycoSwitch® please note that this is a pre-launch product offering. The strains and vectors are offered with supporting information available from BioGrammatics upon request. Alternative reagents or vectors are also available from Life Technologies. The Pichia GlycoSwitch® strains, SuperMan5 and SuperMan5(his-), can be handled similarly to Pichia Classic strains X33 and GS115, respectively.  You can rely on Life Technologies’ Pichia Expression Kit Manual for Expression of Recombinant Proteins in Pichia pastoris (related to Catalog no.     Purchaser Notification Limited Use Label License Pichia Glycoswitch® Expression System
    The Pichia GlycoSwitch® Expression System is based on the yeast Pichia pastoris. Pichia pastoris is an established host for high-level expression of recombinant products. The Pichia GlycoSwitch® Expression System consists of engineered Pichia pastoris strains and compatible vectors. The Pichia GlycoSwitch® SuperMan5 strains and vectors allow for the engineering of human-like glycosylation structures on the protein of interest. Patents, strains, vectors and other materials related to the Pichia GlycoSwitch® Expression System and licenses for its use as to express recombinant products are owned by Research Corporation Technologies, Inc. (RCT), Tucson, Arizona.

    BioGrammatics, Inc. (“BioGrammatics”) has a license from RCT to sell Pichia GlycoSwitch® Expression Systems and related information (collectively, “Expression Kits”) to third-parties for non-commercial research purposes only. A separate license is required for any commercial use, including use of the Pichia GlycoSwitch® strains or vectors for research purposes in product development, product production and/or product improvement. Before using any Pichia GlycoSwitch® strains and vectors, please read the license agreement described below. If you agree to be bound by its terms, please click on the “I agree” box on this order form.

    BioGrammatics grants you a non-exclusive license to use the enclosed Pichia GlycoSwitch® Expression Kit for non-commercial research and evaluation purposes only. The Expression Kit is being transferred to you in furtherance of, and reliance on, such limited use license. You may not use the Expression Kit for any commercial purpose without a license for such purpose from Research Corporation Technologies, Inc. (RCT), Tucson, Arizona. Commercial purposes include: any use of the Expression Kit or Expression Products (as defined below) in a Commercial Product (as defined below); any use of the Expression Kit or Expression Products in the manufacture of a Commercial Product; any sale or other commercialization of Expression Products; any use of the Expression Kit or Expression Products to facilitate or advance research or development directed to a Commercial Product; and any use of the Expression Kit or Expression Products to facilitate or advance any research or development program the results of which will be directly applied to the clinical development or manufacture of a Commercial Product. “Expression Products” means products expressed with the use of the Expression Kit, or with the use of any Pichia GlycoSwitch® strains or expression vectors. “Commercial Product” means any product intended for sale or commercial use. Commercial entities and non-profit entities engaged in commercial activities may conduct their research and evaluation for one year at which time this license automatically terminates. Commercial entities and non-profit entities engaged in commercial activities will be contacted by RCT during the evaluation period regarding their desire for a commercial license.

    Access to the Expression Kit, Pichia GlycoSwitch® strains and expression vectors must be limited solely to those officers, employees and students of your entity who need access to perform the above-described research or evaluation. You must inform each such officer, employee and student of the provisions of this license agreement and require them to agree, in writing, to be bound by the provisions of this license agreement. You may not distribute the Expression Kit or any Pichia GlycoSwitch® strain contained herein or in the Expression Kit (or any derivative thereof), to others, even those within your own entity. You may only transfer modified, altered, or original Pichia GlycoSwitch® strains or material from the Expression Kit to a third party following written notification of, and written approval from, BioGrammatics and RCT so that the recipient can be properly licensed. You may not assign, sublicense, rent, lease or otherwise transfer this license agreement or any of the rights or obligations hereunder, except as expressly permitted by BioGrammatics and RCT.

    This license agreement is effective until terminated. You may terminate it at any time by destroying all Pichia GlycoSwitch® strains, vectors and Expression Products in your control. It will also terminate automatically if you fail to comply with the terms and conditions of the license agreement. You shall, upon termination of the license agreement, destroy all Pichia GlycoSwitch® strains, vectors and Expression Products in your control, and so notify BioGrammatics in writing.
